Several League of Legends champions in Arcane were already playable before the show existed; others became champions afterwards, which is why the overlap keeps growing.

The six leads

Vi, Jinx, Jayce, Caitlyn, Viktor and Ekko carry the story across both seasons and were established champions well before Arcane aired.

Champions playing supporting roles

Heimerdinger — Piltover council member, present from season one.
Singed — chief scientist, working first for Silco and later Ambessa.
Warwick — the transformation of Vander, Vi and Jinx's adoptive father, across the season one timeskip.
Renata Glasc — part of Piltover and Zaun's power struggle.

Champions the show introduced first

Two characters central to season two joined the playable roster around the show rather than before it:

Mel Medarda — council member, Ambessa's daughter.
Ambessa Medarda — the Noxian warlord driving much of season two's conflict.

Who's not a champion

Silco and Vander are major characters but remain show-only — Riot has not turned either into a playable champion, so do not go looking for them in the client.
